An aligned fibrous and thermosensitive hyaluronic acid-puramatrix interpenetrating polymer network hydrogel with mechanical properties adjusted for neural tissue

Abstract Central nervous system (CNS) injuries such as stroke or trauma can lead to long-lasting disability, and there is no currently accepted treatment regenerate functional CNS tissue after injury. Hydrogels mimic the neural extracellular matrix by providing a suitable 3D structure mechanical properties have shown great promise in regeneration. Here we present successful synthesis of thermosensitive hyaluronic acid-RADA 16 (Puramatrix™) peptide interpenetrating network (IPN) that be applied situ injection. Thermosensitive acid (HA) was first synthesized combining HA with poly( N -isopropylacrylamide). Then, Puramatrix™ self-assembled combined produce series injectable thermoresponsive IPNs. The HA-Puramatrix™ IPNs formed hydrogels successfully at physiological temperature. Characterization SEM, rheological measurements, enzymatic degradation swelling tests performed select IPN optimized for neurologic use. SEM images dry demonstrated an aligned porous structure, measurements showed were elastic, elastic modulus approximately 500 Pa, similar brain tissue. An evaluation cell–material interactions also had biological characteristics required engineering, strongly suggesting hydrogel possessed beneficial regeneration
